BackgroundIsoform 1 acts as an extracellular chaperone, preventing nonnative protein aggregation. It stops stress-induced aggregation of blood plasma proteins. It also inhibits amyloid fibril formation by various proteins, including APP, APOC2, B2M, CALCA, CSN3, and SNCA, as well as aggregation-prone LYZ variants (in vitro). It doesn't need ATP and maintains partially unfolded proteins in a suitable state for other chaperones like HSPA8/HSC70 to refold. This isoform doesn't refold proteins independently. Interaction with cell surface receptors leads to internalization of the chaperone-client complex, followed by lysosomal or proteasomal degradation. Secreted isoform 1 protects against apoptosis and complement-induced cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ubiquitin-protein ligase complexes, promoting target protein ubiquitination and proteasomal degradation. They facilitate the degradation of COMMD1 and IKBKB. Modulation of NF-kappa-B transcriptional activity occurs. Nuclear isoforms trigger apoptosis, while mitochondrial ones inhibit cytochrome c release and apoptosis. The protein also plays a role in regulating cell proliferation.
