BackgroundFunctions as a stress-responsive transcription factor, orchestrating the heat shock response (HSR) by activating heat shock proteins (HSPs) that safeguard cells from stress-induced damage. In unstressed cells, resides in an HSP90-containing complex, remaining inactive. Upon stress, it trimerizes, binds specific heat shock elements (HSEs) in HSP gene promoters, and activates their transcription. This process is reversible. Binds chromatin at heat shock gene promoters and has non-transcriptional roles too. Represses Ras-induced c-fos transcription during heat stress, aids HSP70 mRNA processing and export, regulates mitotic progression, and negatively influences DNA repair. Facilitates stress-driven cancer cell proliferation through IER5 dependence.
