BackgroundTyrosine-protein kinase that functions as a cell-surface receptor for PDGFB, PDGFD, PDGFA, and PDGFB/PDGFA heterodimers. It plays a critical role in embryonic development, regulating processes like proliferation, differentiation, survival, chemotaxis, and migration. It's vital for blood vessel development, pericyte and smooth muscle cell recruitment, and normal cardiovascular and kidney glomerulus development. This kinase's activation by various ligands leads to diverse signaling cascades, modulated by interactions between PDGFRA and PDGFRB. It phosphorylates several proteins including PLCG1, PIK3R1, PTPN11, RASA1/GAP, CBL, SHC1, NCK1, SRC kinases, and others. These signals mediate cellular responses such as actin rearrangement, calcium mobilization, protein kinase activation, and cytoskeletal changes. Receptor signaling is regulated by dephosphorylation and internalization.
